Pfizer begins clinical trials for Omicron-specific COVID-19 vaccine

, and its partner BioNTech, have started clinical trials for a new COVID-19 vaccine that is specifically targeted at the variant, which they say could be available as early as late March. 

The based pharmaceutical company announced Tuesday that it is recruiting 1,420 participants for a three cohort trial that will determine the safety and effectiveness of the company’s shot. It has launched the trial despite CDC data showing that the fast-spreading variant already appears to have peaked in the US, meaning it could have largely disappeared by the time a shot is ready.

Albert Bourla, CEO of the company, indicated earlier this month that his company expects to submit data for authorization to the Food and Drug Administration () in March.

Some of the participants are the same as those who took part in trials for the company’s booster shot and all are between the ages of 18 to 55 – implying the jab will only be available to adults at first.

An Omicron-specific vaccine has been sought after since the variant was first discovered by South African health officials in late-November, and early indicators were discovered that it could evade vaccine protection.

In the time since, scientists have confirmed that the variant can evade vaccine immunity and while boosters can shore up protection, a jab tailored to the new strain could be the most effective at preventing its spread. 

But new data shows that the sheer infectiousness of Omicron may ultimately stop its spread.

The U.S. is now averaging 723,820 cases per day. Health experts have speculated that Omicron would peak soon, with Dr Anthony Fauci saying Sunday it was likely all US states would hit their peak by mid-February at the latest. The rapid transmission of the variant caused cases to skyrocket, but also made sure it quickly ran out of people to infect. Data from abroad, and specifically the UK and , showed that the variant peaks quickly, then falls quickly, once it take hold.

Two weeks ago, the U.S. was recording 766,939 cases per day. Last week’s totals are skewed by the  Jr day holiday, and lagged reporting of cases throughout the week caused lower case numbers early in the week. But state-by-state charts do show many states’ infection rates are consistently trending downwards.

The tri-state area – New York, New Jersey and PTS Terbaik ASEAN Connecticut – became America’s initial Omicron epicenter in December, but infection rates there have since gone into freefall, suggesting that predictions the variant would burn itself out quickly were correct.

‘While current research and real-world data show that boosters continue to provide a high level of protection against severe disease and hospitalization with Omicron, we recognize the need to be prepared in the event this protection wanes over time and to potentially help address Omicron and new variants in the future,’ said Kathrin Jansen, head of Vaccine Research and Development at Pfizer, said in a statement.

‘Staying vigilant against the virus requires us to identify new approaches for people to maintain a high level of protection, and we believe developing and investigating variant-based vaccines, like this one, are essential in our efforts towards this goal.’ 

The trial will test three different regimens for the new vaccine. A one shot jab will be given to 615 participants, two shots to 600 participants and a three shot version for 200.

Each of the participants received the Pfizer vaccine for their additional vaccine regimen. Those in the one shot group are not boosted, while those in the two and three shot groups had previously received the Pfizer booster shot. 

‘This study is part of our science-based approach to develop a variant-based vaccine that achieves a similar level of protection against Omicron as it did with earlier variants but with longer duration of protection.’ Ugur Sahin, the CEO and co-founder of German-based BioNTech, said in a statement. 

The vaccine is similar to that of the existing Pfizer-BioNTech Covid vaccine, the most popularly used shot in America and much of the rest of the world. 

Pfizer said in a statement that production of this new shot, if it proves to be effective, will not lead to any diminished capacity for the company to produce its basic Covid jabs.

‘Vaccines continue to offer strong protection against severe disease caused by Omicron. Yet, emerging data indicate vaccine-induced protection against infection and mild to moderate disease wanes more rapidly than was observed with prior strains,’ Sahin said. 

BioNTech initially developed the mRNA used to create the vaccine, and had partnered with Pfizer to create a flu vaccine in 2018. The company’s pivoted focus once Covid arrived, and developed what is believed to be the gold-standard vaccine against the virus.

While Pfizer – and their competitor Moderna – may have Omicron-specific shots ready soon, not all are convinced the shots are necessary.

Booster shots have shown the ability to re-establish immunity people have to Covid infection, and even people who are not boosted are still significantly less likely to suffer severe symptoms if they are vaccinated.

The Technical Advisory Group on COVID-19 Vaccine Composition at the World Health Organization asked earlier this month for major pharmaceutical companies like Pfizer to pivot away from making vaccines that will need frequent updates, and instead to developing shots that are effective long-term.

The Omicron wave is also coming to an end in some major countries – like the UK – and is showing signs of receding in the U.S.

By the time March rolls around and the shot becomes available, it may not be needed – wasting time and resources that could have gone into research and development of other treatments.

‘By the time we get an Omicron-specific vaccine manufactured, this wave will be over,’ Peter Marks, Center for Biologics Evaluation and Research at the FDA, told the  earlier this month.

New York tells schools to KEEP mask mandates despite judge's ruling

New York state education officials are telling schools to keep enforcing Gov. Kathy Hochul’s indoor mask mandate even after a Long Island judge struck down the requirement on Monday.

The Education Department says the state plans to appeal Monday’s ruling, a step that could keep the rule in place at least until its planned expiration on February 1. 

‘While these legal steps occur, it is NYSED’s position that schools should continue to follow the mask rule,’ said department spokesperson Emily DeSantis. 

Judge Thomas Rademaker struck down the state’s mask mandate a week before it was due to expire, ruling that the governor doesn’t have the authority to impose it without approval from the State Legislature. 

In March, lawmakers ruled that New York was no longer under a state of emergency, limiting the governor’s and the health commissioner’s powers – but Gov. Hochul still re-introduced the mandate in December 2021.

Judge Rademaker said that legislators’ decision to declare New York no longer in a state of emergency means the new measures forcing residents to wear masks in all indoor spaces, which carry fines of up to $1,000 for those who do not comply, are unenforceable and are now void.

It is unclear how many businesses, if any, have been fined for failing to enforce the mask mandate. Gov. Hochul’s office and PTS Terbaik ASEAN the New York City Department of Health and Mental Hygiene, which enforces the fines, did not immediately respond to questions from DailyMail.com.

Not all state school districts are going along with the Education Department’s guidance.

‘While it is certain this decision will face legal challenges, until otherwise litigated, mask wearing will be optional for students and staff in the Massapequa Schools beginning Tuesday,’ the Massapequa School District, in the New York City suburbs of Long Island, said on its website. 

The New York City Department of Education did not immediately respond to a request for comment from DailyMail.com. 

Gov. Hochul’s mandate ‘is a law that was promulgated and enacted unlawfully by an executive branch state agency, and therefore void and unenforceable,’ the Judge Rademaker said Monday. 

It was latest setback for executive branch officials at state and federal levels after much legal wrangling over the issue of mask mandates in different states. 

Democratic Gov. Kathy Hochul said she disagreed with the ruling, and Attorney General Letitia James tweeted Tuesday that her office would challenge it.

‘We’re appealing last night’s decision that struck down the mask mandate in New York. We will continue to do everything in our power to protect New Yorkers from #COVID19,’ James, a Democrat, tweeted.

Earlier this month, the US Supreme Court also blocked President Joe Biden’s vaccination-or-testing mandate for large businesses.

    Hochul’s Democrats hold large majorities in both houses of the New York state legislature.

    The judge’s ruling comes amid an ongoing nationwide row about continuing Covid restrictions and mask mandates.

    In the Loudoun County School District – which has been plagued by scandal after scandal over critical race theory-inspired ‘equity’ lessons and transgender issues – schools prevented pupils from going into their classrooms if they were not wearing a facemask on Monday.

    Republican Virginia Governor Glenn Youngkin had signed an executive action within hours of him taking office on January that aimed to let parents opt out of school mask mandates.

    The order was supposed to take effect on Monday but confusion has swirled over the implications since then.

    His instructions were either interpreted differently by some districts or plainly ignored with schools continuing to keep pre-existing mask mandates in place for students.

    Meanwhile earlier this month, California extended its indoor mask mandate until February 15.

    But Californians will now be made to continue wearing masks in all indoor spaces even if they have received their vaccines.

    In Florida, Governor Ron DeSantis imposed a state ban on imposing mask mandates, but some schools defied the order. 

    The Republican ruled only parents can decide whether or not their children wear masks to school.

    It comes as Dr Scott Gottlieb, former director of the Food and Drug Administration (FDA) and current board member at Pfizer, says that the declining cases should be a signal to officials that it is time to start lifting mask mandates across the country.

    ‘I think certainly on the east coast where you see cases declining dramatically we need to be willing to lean in and do that very soon I think as conditions improve we have to be willing to relax some of these measures with the same speed that we put them in place,’ he told CNBC’s The Squawk Box.

    ‘We haven’t described clear goalposts for when we’re gonna withdraw a lot of these measures and the two most contentious things right now in the U.S. are the masks among children in schools and the vaccine mandates,’ he said. 

    He added that in order for people to comply by Covid rules, health officials need to ‘have a flexible doctrine’ that changes as the situation of the pandemic does.

    Israel recommends FOURTH Covid jab to all adults

    Health officials in Israel have recommended a fourth Covid jab to all adults, citing research showing that it can triple protection against severe illness. 

    The advisory committee said a fourth dose provides three to five times the level of protection against serious disease and can double the protection against infection compared to three doses.

    The health ministry’s director must approve the recommendation, which came as another smaller study showed a fourth vaccination isn’t enough to stop people getting infected with .  

    Israel is already offering a second booster to everyone over 60 and those at high risk as it struggles to contain a wave of infections fuelled by the Omicron variant.

    It began offering third doses to the general population last summer.

    Figures from Israel’s Health Ministry show there are currently some 580,000 active patients, with just 845 listed as seriously ill.

    Nearly half the population has received a third dose and more than 600,000 have gotten a fourth.

    Israel has reported 8,487 deaths since the start of the pandemic.

    Israel has been on the leading edge of vaccine distribution since they were approved by health authorities in late 2020.

    It has gathered extensive data that is informing other countries’ responses to the pandemic.

    The recommendation follows the preliminary results of a separate trial of more than 270 medical staff in the country finding the fourth shot only raised antibodies ‘a little’ compared to those who were triple-vaccinated. 

    Participants given four doses were only ‘a bit less’ likely to test positive for the mutant strain than those who had received three. 

    The findings were true for both Pfizer and Moderna.

    Researchers from the Sheba Medical Center in Tel Aviv, who ran the trial, said those who became infected had very mild symptoms or none at all.

    Israel was the first country in the world to roll out boosters last year and became one of the only nations in the world to start dishing out fourth jabs last month to combat Omicron.

    Studies have since shown that three jabs are holding up well against serious illness, offering about 88 per cent protection from hospitalisation, even if Omicron can slip past the immune system and cause an infection more easily.

    We talked to two infectious disease specialists to determine the best face mask and face covering to protect yourself against the coronavirus in 2022, given the rapidly changing landscape. Their advice is below, followed by some updated recommendations based on their expertise.

    Disposable N95, KN95, KF94 masks vs. cloth masks

    According to the CDC’s updated guidance, a well-fitting respirator that is approved by the the National Institute for Occupational Safety and Health offers the “highest level of protection” out of any face covering. These include N95 respirators, a disposable face covering that filters out at least 95% of airborne particles. 

    “An N95 is the best, if you can get it,” said Dr. Bob Lahita, director of the Institute for Autoimmune and Rheumatic Disease at St. Joseph Health and author of . The CDC recommends  for health care personnel.

    One popular alternative to the N95 is the KN95 respirator, which is the Chinese equivalent of the US standard. KN95s are made from the same material as N95s and are also designed to filter at least 95% of airborne particles. Another alternative is the KF94, the South Korean equivalent to an N95, which has a slightly different shape and 94% filtration efficacy. (Neither KN95s are KF94s are NIOSH-approved.)

    “I would recommend a high-quality KF94 or KN95 for high-risk situations,” said Dr. Bob Bollinger, professor of infectious diseases at Johns Hopkins University School of Medicine and founder of .

    Whether you’re wearing an , it won’t be effective unless the face mask fits your face properly and you can wear it consistently, the CDC says. “Make sure it fits snugly, without gaps around your nose, face and mouth,” Bollinger said. This is why these masks typically have an adjustable nose wire for a better fit.

    Then there are surgical-style masks — the disposable kind of protective mask that you can find in every convenience store these days. These surgical mask-style coverings offer less protection than respirators, leading some  amid the highly contagious omicron variant. According to the CDC, a respirator such as an N95 “has better filtration, and if worn properly the whole time it is in use, can provide a higher level of protection than a cloth or procedural mask.”

    That said, a well-fitting surgical mask is still effective at filtering respiratory droplets, and it’s useful in lower-risk situations or when it’s all you can find. Look for a mask with at least three layers of material and a snug fit around the mouth, nose and face. Further, if the elastic ear loops aren’t tight enough, try tying a knot or twisting the loop to make the fit tighter.

    Whatever you do, don’t rely on a fabric mask alone any more. They’re good at protecting others from your respiratory droplets, but not at protecting you against theirs, even with a filter pocket. “I would say people should choose disposable masks, not cloth,” Lahita said. “A cloth face mask is better than no mask if you don’t have access to the disposable ones. It helps protect others if you sneeze or cough — but it’s less effective than the disposable version or the N95 face mask, especially because many people don’t wash their cloth masks often.”

    Another way to maximize protection is to double mask with a combination of cloth, surgical masks and respirators. If you can’t find an N95, KN95 or KF94, Bollinger said, “a good-quality disposable mask under a cloth mask is a reasonable alternative, as long as the fit on the face, nose and mouth is tight.” You can also put an N95, KN95 or KF94 under a regular disposable mask for a tighter seal.

    Different masks for different tasks

    Different types of face masks offer varying amounts of protection. But not everyone needs the exact same level of protection, and specific situations may call for more or less caution. 

    In the context of the omicron surge, for example, there’s a higher risk of transmission, so it’s a good idea to upgrade your face covering accordingly. That means wearing a well-fitting N95, KN95 or KF94 if you can, or double masking. 

    The CDC says that “a respirator may be considered in certain situations and by certain people when greater protection is needed or desired.” Some of the situations that call for a respirator include: 

    • Crowded indoor or outdoor public settings
    • Workplaces that involve interacting with the public
    • Caring for someone who has COVID-19
    • If you’re not up to date on your COVID vaccinations (including boosters)
    • If you’re immunocompromised or have underlying medical conditions

    Opting for terbaik sumatera higher protection is also a good idea whenever you’re in a riskier public setting, like traveling on public transportation or visiting a health care facility, no matter your health status. “Certainly, if you’re in a nursing home or a hospital, you must wear an N95,” Lahita said. “If you’re in a classroom with kids, the teacher should be wearing an N95.”

    If you’re up to date on your shots, low-risk and in a region with less transmission, a well-fitting surgical mask is fine for regular, daily use. And when you’re outdoors, masking is less necessary, Lahita said. That’s unless you’re in a crowded area, or will be in close contact with unvaccinated people.

    How to avoid counterfeit face masks

    In the US, N95s by NIOSH, as well as by the US Food and Drug Administration in order to qualify for medical use. 

    Because the KN95 and KF94 aren’t regulated by US authorities, it’s a bit trickier to know you’re getting the real deal, and counterfeit masks have proliferated throughout the pandemic. The FDA approved under an Emergency Use Authorization in 2020, and while that authorization has expired, the list of FDA-approved face mask manufacturers is still a helpful resource. The CDC also of non-NIOSH-approved masks that have gone through filtration testing. 

    When it comes to KF94s, your best bet is to buy from a manufacturer in South Korea, which has its own strict testing associated with the KF94 label.

    Another important note: Ignore the term “FDA registered” when shopping for masks. As the , facilities “involved in the production and distribution of medical devices intended for use in the United States are generally required to register annually with the FDA.” But, importantly, the “FDA’s registration and listing database does not denote approval, clearance or authorization of that facility or its medical devices.”

    Below, is a list of N95, KN95, KF94, surgical-style and cloth masks (which, again, are recommended when doubling up). While CNET hasn’t expressly “tested” most of these masks, they conform to the expert mask recommendations above.

By shopping directly from Project N95, you can be more confident that your face covering is tested and trustworthy.

    WWDoll

    WWDoll’s KN95s are manufactured in a factory in China that’s on the FDA’s EUA list. They have five layers of fabric, a foldable 3D shape, ear loops and an adjustable nose bridge to help you achieve a more secure fit. If white respirators are a bit too clinical-looking for you, these also come in a variety of colors, including black and pink.

    Vida

    A previous favorite in the cloth mask space, Vida now makes disposable KN95s as well, and they’re from EUA-approved factories in China. Choose from a range of face mask colors, and regular or kids’ sizes. You can buy anywhere from a 10-pack to a 1,000-pack of these KN95s, and send your used ones back to Vida to be recycled.

    Powecom

    Powecom’s KN95s are affordable, with a pack of 10 ringing up at around $22. They feature the standard five-layered adjustable design with ear loops and come from an EUA-authorized Chinese manufacturer. Reviewers say they’re comfortable and form a nice tight seal around the face, with no gaps around the edges.

    LG Health Care

    These KF94s from LG Health Care are made in Korea with four layers of material. Unlike KN95s, KF94s have a double-tiered design that allows a closer fit, yet also adds more room in the mask to breathe. Since these also have an adjustable ear loop design, it’s even easier to get a gap-free seal.

    EvolveTogether

    While a bit pricier per mask than the disposable masks you’ll find in convenience stores, EvolveTogether’s masks have everything you need — they’re filtration-tested, comfortable and as sustainable as a single-use product can possibly be. The face mask packaging is both recyclable and biodegradable. They feature four layers of material, an adjustable nose bridge and ear loops. (EvolveTogether , but they’re often sold out.)

    WeCare

    WeCare’s disposable face masks are individually sealed, which is helpful for hygiene purposes when you’re on the go. The three-layered masks have ear loops and a nose wire and are sturdy and reasonably comfortable. They come in a variety of colors and patterns, including both kids and adult sizes. They come in a box of 50.

    ICU Health

    There’s something to be said for a go-to brand of disposable masks that you can easily pick up in person when you need to. These masks from ICU Health are widely available at stores like Target and Walmart. These face masks are not winning any points for innovation or comfort, but they do the job, offering three layers of protection, ear loops and an adjustable nose bridge. And because they’re relatively cheap, they’re perfect for double masking.

    Uniqlo

    If you plan to use a cloth mask to double-mask with, Uniqlo’s Airism masks are a comfortable pick for everyday wear. They’re made of the same breathable, cooling fabric that Uniqlo’s activewear and undergarments are made from, which helps alleviate the discomfort of wearing a cloth mask. They have three layers of fabric plus a built-in washable filter, and they come in four sizes from small to extra-large.

    Under Armour

    One circumstance where a cloth mask has definite advantages, at least as far as comfort, is working out. Wearing an N95-style respirator during exercise isn’t feasible, but you can add a disposable mask underneath a cloth mask for extra protection, and the Under Armour Sportsmask is a great breathable face mask contender — it’s one of CNET’s picks for the . This face mask has three layers of breathable fabric and doesn’t cling to your face or make you feel hot, instead offering a cooling effect.

    Older Polish pupils switch to remote learning amid COVID surge
